Kinoxis Therapeutics Pty Ltd is a private, Australian-based, clinical-stage biotechnology company developing first-in-class therapeutics targeting ALOX15, a key regulator of neuroinflammation and neural excitability in the brain. By modulating disease-relevant biological pathways, Kinoxis is advancing mechanism-based precision therapies to address significant unmet medical needs in neuropsychiatric and central nervous system disorders. Kinoxis’ lead clinical candidate, KNX100, is a first-in-class, short-acting small-molecule inhibitor of ALOX15 being developed for the treatment of acute agitation and aggression associated with dementia and other neuropsychiatric conditions. The compound has demonstrated robust activity across multiple preclinical models and was shown to be safe and well tolerated in a completed Phase 1 clinical study conducted under a U.S. Investigational New Drug (IND) application. KNX100 is currently being evaluated in the CARES-X Phase 2 clinical trial, a multi-site study in Australia assessing its safety, tolerability, and efficacy in patients with agitation associated with dementia. In parallel, Kinoxis is advancing KNX101, a second-generation, long-acting ALOX15 inhibitor designed to address acute and chronic pain and chronic agitation through sustained modulation of neuroinflammatory signalling pathways. These programs form part of Kinoxis’ broader strategy to deliver novel, non-opioid and non-sedating therapies for patients living with complex neurological and psychiatric conditions.
